چکیده

Bacterial auto-aggregation is a critical step during adhesion of N. meningitidis to host cells. The precise mechanisms and functions of bacterial auto-aggregation still remain to be fully elucidated. In this work, we characterize the role of a meningococcal hypothetical protein, NMB0995/NMC0982, and show that this protein, here denoted NafA, acts as an anti-aggregation factor. NafA was confirmed to be surface exposed and was found to be induced at a late stage of bacterial adherence to epithelial cells. A NafA deficient mutant was hyperpiliated and formed bundles of pili. Further, the mutant displayed increased adherence to epithelial cells when compared to the wild-type strain. In the absence of host cells, the NafA deficient mutant was more aggregative than the wild-type strain. The in vivo role of NafA in sepsis was studied in a murine model of meningococcal disease. Challenge with the NafA deficient mutant resulted in lower bacteremia levels and mortality when compared to the wild-type strain. The present study reveals that meningococcal NafA is an anti-aggregation factor with strong impact on the disease outcome. These data also suggest that appropriate bacterial auto-aggregation is controlled by both aggregation and anti-aggregation factors during Neisseria infection in vivo.

Colony morphology of piliated Neisseria meningitidis

An association between piliation and colony morphology has not been observed for the meningococcus. We have found that growth of meningococci overnight at 30 degrees C in a candle extinction jar allows observation of distinct colonial phenotypes correlated to the presence or absence of piliation and the expression of opacity-associated proteins. These phenotypes are similar to those observed in...

Packaged identification systems and bacteria of veterinary origin.

Thirdly, this isolate was more resistant to spectinomycin than what is generally experienced with N. gonorrhoeae cultures. Our laboratory tests approximately 3,500 N. gonorrhoeae cultures annually, and the MIC of spectinomycin is higher for N. meningitidis than for N. gonorrhoeae (3). It is of particular interest that a second similar microorganism has been isolated in Canada from a female geni...

The meningococcal PilT protein is required for induction of intimate attachment to epithelial cells following pilus-mediated adhesion.

The ability of Neisseria meningitidis (MC) to interact with cellular barriers is essential to its pathogenesis. With epithelial cells, this process has been modeled in two steps. The initial stage of localized adherence is mediated by bacterial pili. After this phase, MC disperse and lose piliation, thus leading to a diffuse adherence. At this stage, microvilli have disappeared, and MC interact...

ComE, a competence protein from Neisseria gonorrhoeae with DNA-binding activity.

Neisseria gonorrhoeae is naturally able to take up exogenous DNA and undergo genetic transformation. This ability correlates with the presence of functional type IV pili, and uptake of DNA is dependent on the presence of a specific 10-bp sequence. Among the known competence factors in N. gonorrhoeae, none has been shown to interact with the incoming DNA. Here we describe ComE, a DNA-binding pro...

The number of Neisseria meningitidis type IV pili determines host cell interaction.

As mediators of adhesion, autoaggregation and bacteria-induced plasma membrane reorganization, type IV pili are at the heart of Neisseria meningitidis infection. Previous studies have proposed that two minor pilins, PilV and PilX, are displayed along the pilus structure and play a direct role in mediating these effects. In contrast with this hypothesis, combining imaging and biochemical approac...
